1. Add in 1 mL of 2000 Dextran blue.

2. Let it flow through the bed until all the Dextran blue was within the chromatographic bead before adding in dH2O. This is to prevent dilution of the Dextran blue

3. let it flow until the blue color reaches the bottom of the column.

4. The volume collected were measured with a cylinder to obtain the void volume.

Packing Chromatography column

1. weigh 3g of chromatography beads (for 50 mL of column) --> this depends on the size of the column. the beads usually swell 5-6 times its original size after heating.

2. Add in distilled water and starts stirring it with magnetic stirrer and heating it at 60 celcius. Discard the impurities on the upper layer once in a while until the mixture was free of impurities.

3. Incubate in the oven overnight for full swelling.

4. The bottom of the column was stuffed with cotton.

5. The swollen beads was poured into the column slowly.

6. The screw of the cork at the bottom of the column was opened to release water (slowly for so that the beads would pack better)

7. When the column was more than half packed, the screw of the cork was opened to its maximum.

8. While packing the wall of the column should be knock gently to allow uniform packing.

Lessons learnt

1. Chromatography beads are very expensive. It is recyclable. Even if the column cracked during packing, it could still be repacked. Never discard it.

2. Do not start doing things without consulting your senior

3. Chromatography beads have to be kept in the fridge (4 celcius) after recollecting it. It should be immersed in distilled water(or its buffer).

4. Proteins: if are to be left for more than a day has to be kept in the fridge.

5. Tissue papers, gloves, and noodle cups go into the waste paper basket and not the biohazard plastic bag.

6. After centrifugation, pour the supernatant in one go.

I'm just too pissed off with people who are still distributing fansubs although they had promised not to do so when they join the community!!! They're making everyone's life harder and harder. Dramacrazy, huh? stealing other people's hard work and they still have the nerve to mock the fansubbers! ArrgGGHhhh~~!! DAMN IT!

I had my panda eyes on today because i cried a lot yesterday... I happened to watch a raw video of Tensai Shimura Doubutsuen (2008.04.12) last night. Only then that i realize that the movie of "A tale of Mari and Three Puppies" was based on a true story. I searched high and low for the download links / torrent for the movie but failed. Instead i found a blog (http://redstarcafe.wordpress.com/2008/01/26/mari-and-the-niigata-earthquake/) which gives a complete summary of the movie. Read it, watched the trailer and cried again.

How to Control Your Temper

1. Remember that the reward for patience is happiness.
2. Realize that things can be handled well at all times; it's just a matter of what kind of attitude you put into it. Take deep breaths.
3. Get away from situations where your anger might get the better of you, such as when someone is teasing you. Ignore the other person and walk away. If you must, do this a few times until the person gets the hint that you will not tolerate it. It's okay to use your words or body language to tell somebody you will not tolerate how they are treating you, as long as you respond to them in a calm and matter of fact manner, without being overtly hurtful or trying to embarrass them as they have you. It's never okay to hit somebody for making fun.
4. Realize that no one likes to be around people who get angry easily, and act abusive. The only way you can change the situation is by starting with yourself.
5. Be considerate of others, and ultimately, yourself. The way you react will ultimately affect your surroundings. If you have to rebuke someone for doing something wrong, you can always do it calmly, in a private place. This method is always better for everyone in the long run.
6. If you are angry and you are standing, sit down, if you are sitting then lie down.
7. Remember, 5 seconds stand between you and logic, count to 5, and remember that logic always beats anger.
8. Stop talking to that person for few minutes to whom you are angry and take deep breath around 5-7 times.
9. (IF there are any) Think about the good things that person has done for you. What bad have you done to them? Chances are, you've done at least something to them to make them resentful towards you. Make sure you have a clear conscience-then speak.
10. Temper can ruin anybody's life. When your eyes are opened it might be too late, so try to control your anger and follow the above steps. It will definitely get you away from tension.
11. Count from 1 to 10 in ascending order and then descending order, you will feel a bit calmer.
12. Saying hurtful things could be very risky and dangerous and it can hurt someone's heart so please stay calm when you feel hyper.

And i think this is very useful especially for someone like me.... I really find it difficult to control my temper... Sometimes, because of my temper, i'm driving people away... T___T
I get angry very easily but i would regret very soon too... this is bad....

Never have i thought finding a job is more difficult than doing my FYP + thesis..
My friend attended an interview and it was so stressful that my friend's confidence went rupturing. The interviewers did not ask anything related to your field. In stead questions on your common sense were asked. Their main objective was to attack the candidates' self-confidence and to observe the choices and decisions the candidates were making.

Over 500 people sent their resumes, but only 70+ people were selected to undergo the first phase of the interview. The first phase only consisted of some easy questions and psychological tests. So it was pretty easy. From the first phase, only 20+ people were selected for the second phase. Second phase of the interview consisted of a "so-call game". 4 person would be grouped together to solve group assignments.
Questions like this would be asked:

"If there were 8 people (doctor, an old woman, engineer etc etc) in a flight that is about to crash, and there were only 7 parachutes left, who's supposed to sacrifice him/herself and why?"

The interviewers will not accept answers like "2 person sharing one parachute"...
Group members would be fighting among themselves to answer to gain a "good impression". (Imagine the pressure here - you have to be fast and logical in thinking). There was no specific answer for any problems given as long as your answers are logical. Sounds easy right? BUT... all the answers given by the candidates were doubted and countered by the interviewers. Not even one person's answer was acknowledged. At this point, the candidates' confidence would be slowly crushed to the extend where they felt like giving up in the middle.

One good news was my friend successfully went through the interview and was one of the lucky 10 who was offered for the post. I should say she was STRONG and CAPABLE.
